If you want to see the inside of the Sistine Chapel, my advide would be to arrive very early as the line can easily wrap around two blocks very quickly.

The Vatican museums were not (thankfully) on our absolutely-positively-must-see list. St. Peter's Basilica and the Scavi (excavation) tour were. On our first (full) day in Rome, we got to the Vatican fairly early (before 8:30 a.m.) and there was NO line to enter the Basilica. We were able to take our time and see and appreciate everything without waiting for others to make a little room, or worry about holding someone else up. By the time we were finished it was time to pick up our Scavi Tour tickets for our 9:15 a.m. tour. By the time THAT was over and we re-entered the square, the line to enter the Basilica was really lo-o-o-o-o-ong.

On a side note, if you are an art lover, make sure that you go into st. peters basilica to view the Pieta. And you would not complete a trip to Rome without viewing The Origin Of Man on the ceiling of the Sistine Chapel. (word of caution here. Don't speak but in a whisper and, under no circumstances do you try to photograph the ceiling or you will be escorted to the nearest exit) Finally, if you want to see a beautiful art museum, go to the Borghese. You have to have reservations to go inside. You can purchase them and print them out on the internet.
