Is it better to take this tour or do 2 shorter tours on each day. Has anyone done either of these options?

It depends on the where the tour goes you will probably have several tours to choose from.
On the cruise we were on with Orient lines we had 17 different excursions for 2 days

Also depend on your stamina
You may want to check if others on your cruise are doing private tours ..less people about the same cost as ship's tours.
No waiting & wasting time for everyone to be back on the bus at each stop.

Thanks for replying LHT28. Yes we have over 3 pages of tours to choose from. The 2 day combined tour seems to cover almost everything listed on the other tours, but it is expensive. I just wonder if it will be too strenuous over 2 long days. Will check out private tours - good advice - thank you.

It is a long 2 days no matter how you do it.
I found it tiring but in a good way ...we got to see most of the main sights but time was cut short by a couple of ladies on the bus that insisted in wandering off at each stop then we had to wait 10-15 while the guide went to find them. Smaller tours are better with 8 people you can remember who was on your tour & drag them back
